| Description: | Genes of human CYP11B2 and rat NADPH-P450 reductase were transfected into hamster V79 cells using a lentivirus vector, and stably-expressing cell lines were selected by taking advantages of the associated puromycin-resistance gene. Cultured cells were diluted and seeded in a 96-well plate with an amount of approximately 10,000 per well. After pre-incubation with suitable concentrations of the test compounds for 2 hours, 11-deoxycorticosterone of 200 nM was added as the substrate to initiate the catalytic reaction. After further incubation for 30 minutes, it was quenched with ethyl acetate. Subsequently, the substrate and product(s) were quantified using LC-MS/MS or HPLC at 260 nm, and the IC50 values were calculated according to inhibitory percentage. |
