Description:  Final Assay Conditions:Total reaction volume: 25 μlAssay buffer: 100 HEPES pH 7.4, 200 mM NaCl, 2 mM CaCl2, 5 mM DTT35 nM recombinant human PAD4500 μM TSTGGRQGSHH peptide1.2% DMSOStop solution: 10% formic acidReaction mixtures were incubated at room temperature for 30 minutes. 10 μl each of the reaction mixtures was then mixed with 40 μl of 10% formic acid in a microtiter plate. The plate was frozen at −80° C. before shipping out on dry ice for RapidFire mass spectroscopy analysis.Thawed samples were loaded onto the Rapid Fire 300 system (Agilent) wherein they were first sipped for 250 ms and then loaded onto a Agilent “C” (C18) cartridge using a mobile phase of water containing 0.09% formic acid/0.01% trifluoroacetic acid for 3000 ms desalting flowing at a rate of 1.5 ml/min. Once the samples were loaded and washed, a mobile phase of acetonitrile containing 0.09% formic acid/0.01% trifluoroacetic acid was used to elute the samples directly onto a Sciex API4000 triple quadrupole mass spectrometer for 3000 ms at a flow rate of 1.25 ml/min.
