| Description: | The inhibitory activity of the compound for SIK1, SIK2, and SIK3 enzymes was assayed by measuring the amount of ADP, an enzyme reaction product, with ADP-Glo™ Max Assay (Promega). Enzymes, specifically, GST-tagged SIK1 (Carna Biosciences, Inc.), GST-tagged SIK2 (Carna Biosciences, Inc.), and GST-tagged SIK3 (SignalChem) were prepared in a reaction buffer (15 mM tris-hydrochloric acid buffer (pH 7.5), 0.001% Triton X-100, 0.01% BSA) so that the final concentration would be 20 nM, 3 nM, and 100 nM. Enzyme solutions were thus formed. In a reaction buffer, AMARA Peptide (SignalChem) as a substrate with a final concentration of 200 μM (SIK1, SIK2) or 100 μM (SIK3), ATP (Promega) with a final concentration of 1 mM, and magnesium chloride with a final concentration of 5 mM were prepared. Substrate solutions were thus formed. The enzyme solution, the substrate solution, and a DMSO solution of a concentration of the test compound were mixed together and reacted at room temperature for 2 hours. After the reaction, ADP-Glo™ Reagent (Promega) was added, and the reaction was performed at room temperature for 30 minutes. Subsequently, ADP-Glo™ Max Detection Reagent (Promega) was added, and the reaction was performed at room temperature for 30 minutes. The emission by the luciferase reaction was measured with PHERAstar FSX (BMG LABTECH). |
