| Description: | TYK2 (h), together with 8 mM MOPS, pH 7.0, 0.2 mM EDTA, 250 μM GGMEDIYFEFMGGKKK, 10 mM magnesium acetate and [γ-33P]-ATP (activity and concentration of which were tailored as needed) were incubated. Mg/ATP mixture was added to start the reaction. After incubation at room temperature for 40 minutes, 0.5% phosphoric acid was added to terminate the reaction. Then 10 μL of the reactant was dripped on a P30 filter pad, washed with 0.425% phosphoric acid for three times and then with methanol for one time within 4 minutes, dried and counted by scintillation. |
