| Description: | The Gai biosensor consists of a Rluc8-tagged Gαi2 subunit, a GFP10-tagged Gγ2 subunit, and an untagged Gβ1. Agonist stimulation and GPR84 activation triggers a physical separation between the RLuc8-Gai donor and the GFP10-Gγ2 acceptor, resulting in a decrease in BRET signal whose amplitude is correlated to ligand efficacy (Gales et al., 2006). Moreover, signaling functions of GPCRs are tightly regulated by endocytosis, the targeting of receptors to endosomes and their sorting to lysosomes or recycling to the plasma membrane. The early endosomes (EEs) trafficking assay (Namkung et al., 2016. Nat Commun. 7, 12178) uses Rluc8-tagged GPR84 and Renilla GFP (rGFP) attached to the FYVE domain from human endofin/zinc finger FYVE domain-containing protein 16, which binds phosphatidylinositol 3-phosphate in EEs. |
